Users specify if they want comments?

Is it possible to let users choose, if they want to receive comments about their ads on a ad to ad basis?

No this is not an option. You can only set globally via admin edit categories which would be for all ads

I've just recently purchased Classifieds so I haven't had the chance to install and test everything. Is it possible to lock certain discussions if they get out of hand? Can Moderators be appointed?

You can set in the category permissions that users can not comment in that category but there is no specific option to lock a thread since this is not a forum product it does not have forum specific features.

The product is designed around ads not discussions.
